The boys will play two of the leading lady's younger siblings.
The French romantic opera concerns the tragic story of Werther, a man helplessly in love with Charlotte, who is already promised in marriage to someone else.
World renowned mezzo-soprano, Alice Coote, plays Charlotte, and Paul Nilon is Werther. The production is conducted by Opera North's musical director Richard Farnes.
Following the Leeds performances, the boys will go on tour with Opera North to Salford, Nottingham, Belfast, Newcastle and Sheffield.
Andrew Wheeler, head of Instrumental Studies at the Grammar School said: "This is a fantastic musical experience for James and Charles."
The production opens at Leeds Grand theatre on Oct 26.
